0.5 mL of a peptide digest sample is added to 0.5 mL of a 5 mg/mL solution of a-cyanohydroxycinnamic acid solution in 30% acetonitrile in water with 0.1 % TFA. The sample is briefly mixed and 0.5 uL of the mixture is spotted onto a 96 sample MALDI target plate. The plate is inserted into a Applied Biosystems Voyager DE-Pro MALDI-TOF where the digest is examined to assure observation of peptides prior to chromatography.
